The Physicians' Health Study II randomised more than 14,000 male physicians to a daily multivitamin for over a decade and found no reduction in major cardiovascular events. The Cochrane review of antioxidant supplements pooled trials in more than 290,000 participants and found no mortality benefit, with signals of increased mortality for beta-carotene and vitamin E.

None of this says supplements are useless in every case. Trials support replacement where a deficiency is documented, and specific situations such as folic acid in pregnancy or vitamin B12 in people who cannot absorb it. What the evidence does not support is stacking dozens of compounds in a healthy adult to extend lifespan, and there is no trial at all of a regimen of this size, so its combined interactions are untested.
